Why 1ml is used to pour plate method, because there two main reasons: statistical accuracy and practical physics. And By pouring exactly 1 mL, the “Volume Plated” in the denominator becomes 1. This completely eliminates an extra multiplication step. If using more than 1 ml causes agar improper solidification , Less than 1ml leads to sample dryness and might cause death of organisms because of the temperature of the agar . When compared to spread plate this pour plate 1ml volume is 10 times larger than 0.1ml and able to count as per ISO or USP standard guidlines
