Why Salmonella detection only 10 gram sample for MLT Why not 1 gram?

Salmonella is weakest of all pathogens, but in favorable environment it is highly
pathogenic than other. 10 g is worst case scenario. we have to prove it is absent in 10
g sample which is larger quantity used comparing to all other pathogen. USP
requirement Refer chapter 62.


Because we are detecting salmonella as a species not as a single bacteria, So
salmonella spp can not be detected in 1g sample Quantity. Hence large amount of
sample is required to screen the bacteria present in the sample.


Salmonella contamination in pharmaceutical products or raw materials is expected to
be low if present. Testing a larger sample size (10 grams) increases the likelihood of
detecting even small amounts of Salmonella. If only 1 gram were tested, there is a
higher chance that the contamination could be missed.


The testing of 10 grams is in line with regulatory guidelines, such as those provided
by the United States Pharmacopeia (USP), European Pharmacopeia (Ph. Eur.), and
other international standards. These guidelines are designed to provide a robust
method for ensuring product safety by reducing the risk of false negatives.


By testing 10 grams, the statistical confidence in the test results increases, providing
greater assurance that the product is free from contamination.
